Trezeguet couldn't really be a lone striker.

A lone striker has to be able hold the ball up, work to make space and provide a goal threat.

Trez could only provide a goal threat.

Amauri can only hold up the ball.

Iaquinta can only work to create space.

So you are right to say no current Juve striker can effectively as a lone striker.

4-5-1 is not just about defending, but to keep possession because of a spare man in the midfield. and Krasic as usual will be further up the pitch.

A 4-5-1 formation with Melo-Momo-Aquilani looks good. But playing Marchisio out in the left wing again is really starting to annoy me.

I see a lot members here looking excited at the possibility to see Pepe as a left back. Surely, he never gives up, works hard and tracks back and he deserves credit for that, but is he tacticaly smart enough and disciplined to be a defender?
Moreover, he's not a natural defender and i'm not sure if he ever played successfully in that position.

To summarise, at this point a 4-5-1 is not a good idea. We're lacking good players at some crucial positions. Two good full backs, one good left winger and a top centre forward to be precise.
