I believe 4-2-3-1 will be too defensive. Even if Melo and Marchisio plays at the MC position in for example a 4-1-3-2 formation, they will still be quite defensive due to their natural playing style. So I believe Sissoko as the only DM in the formation will do good.

4-2-3-1 ain't really that defensive, 3 offensive midfielders to support the attack, so the 2 DMs are really needed to hold the fort ...

I do not think Sissoko can give his best by placing him as the only anchoring DM, he should be sliding all round the middle of the park, winning the ball for Gio/Diego/DP or make galloping runs at the opponents when he dispossesses them.
