Trémoulinas was born in the commune of Lormont, a suburb located on the outskirts of Bordeaux, and began his career playing for hometown club US Lormont. In 1999, he signed a youth contract with FC Girondins de Bordeaux. Trémoulinas spent approximately seven years in the club's youth system and one year in the Championnat de France amateur before earning a call up to the senior team in preparation for the 2007–08 season following the signing of his professional contract. He was designated the number 28 shirt and served as backup to the younger Florian Marange and elder Franck Jurietti. Trémoulinas made his professional debut on 15 August 2007 in a league match against Le Mans. He was limited to only nine league appearances, but was a regular in the club's 2007–08 UEFA Cup campaign appearing in seven matches and also scoring his first European goal in a 3–2 victory over Greek club Panionios.

Trémoulinas was slowly integrated into the squad for the 2008–09 season by manager Laurent Blanc upending both Marange and Jurietti, though the latter was still relied more upon in important matches, such as when he started for Trémoulinas in matches against Paris Saint-Germain, Lyon, and the 2009 Coupe de la Ligue Final. Trémoulinas did appear in the final as a substitute in the 17th minute for the injured Brazilian Wendel, but had to substituted out at halftime, also due to injury. Bordeaux won the match 4–0 giving Trech, as he is called by the Bordeaux faithful, his first career club honour. Following the final, Trémoulinas started the final five matches of the season helping Bordeaux win its first league title since the year he joined the club. In total with the team, he appeared in 31 matches scoring his only goal of the season against Guingamp in a 4–2 victory in the Coupe de la Ligue.

On 29 May 2009, Trémoulinas was rewarded with a contract extension keeping him with the club until June 2013. Trémoulinas successful play during the 2009–10 Ligue 1 season saw the player attract interest from Italian clubs A.C. Milan, Juventus and Fiorentina, and Spanish club Real Madrid, though Trémoulinas quickly brushed off the rumours. He was among the top assists providers in the league primarily due to his crosses and ability to get forward.

On 5 August 2010, Trémoulinas was called up to the senior team for the first time by new manager Laurent Blanc for the team's friendly match against Norway on 11 August 2010.

    Trémoulinas, the track continues to rise

    Despite the recent denial of the President of the Bordeaux, Benoit Trémoulinas (24) continues to be a target of Juventus. After the statements of a number of the Girondins who had declared that the 24-year old lefty would not move from Bordeaux, Le Parisien trans head arrives to reopen the track that leads outside Lormont: according to these tests, in fact, it seems that left side has been viewed by some emissaries Juventus last Sunday in Ligue 1 competition between Bordeaux and Toulouse. As stated in Le Parisien, Juventus now has not made any formal offer for Trémoulinas but August 31 is still relatively far away ...

    how reliable is Le Parisien? tuttosport of france?

    But I'm not comfortable with two fullbacks who can't defend to save their lives. Same reason why we need DC gone.

    Tremoulinas is surely better than De Ceglie, even when it comes down to defense.

    Left - him and Pepe ; right - Motta and Krasic

    It would be a damn good combination IMO. I just can't see this guy leave France for another year...I hope I'm wrong though.

    I was thinking that was possible as well. Quite weird that the one with better crossing would play in defense though.

    I have no idea about Bordeaux, but he could be tempted to leave now that the messiah went to the national team.

    I know but Bordeaux to let him go in the late August? Not so sure...

    Bordeaux are a sinking ship, he should leave. They already lost their first two games of the season, the last one at home against Toulouse.

    Oh yeah, another thing. Why I also think he's not coming...

    Marotta said that we'll use money for a winger and other deals will be loans and co-ownerships. I don't think we have that much money left.

    MAYBE...maybe if sell Iaquinta. That way Martinez would get to play more. But again, I doubt we can pull it.

    ok. if we had the money for dzeko... why won't we have the money for him? we offered 33million for dzeko.. and krasic deal was 15m..
